THE ONE AND ONLY Drama tells the story of Pyo In Sook, who was left by her parents with her grandmother who raised her when she was young. This has made Pyo In Sook's life never easy and comfortable. As an adult, she works at a public bathhouse to scrub customers' bodies.

And to survive, she never expresses her feelings to anyone. But one day, a doctor tells Pyo In Sook that she has a terminal illness and will soon die. Even though she has heard this, she remains emotionless.

She starts receiving treatment at the hospital and meets two women named Seong Mi Do, a social media influencer, and Kang Se Yeon, an ordinary housewife who feels lonely and is looked down upon. In addition, she also meets Min Woo Cheon, a female assassin.
